Engineering a Human Immunodeficiency Virus (HIV) protein to assist HIV treatment design

The HIV-1 gp160 protein fluctuates among a range of conformations – closed, partially open, and open – exposing different antibody target sites. The “closed” gp160 conformation is a primary target in current vaccine design efforts because it hides poor target sites while exposing conserved target sites. Deep mutational scanning was used to provide insight into gp160 acquiring open and closed conformations; we hope these findings may assist HIV-1 treatment design.
